However, there are growing problems with drug resistance that are posing a threat to the global fight against malaria. Chloroquine is an anti-malaria medicine that works by interfering with the growth of parasites in the red blood cells of the human body.

Hemozoin collects in the digestive vacuole as insoluble crystals. Chloroquine enters the red blood cell, inhabiting parasite cell, and digestive vacuole by simple diffusion. Chloroquine then becomes protonated to CQ2+, as the digestive vacuole is known to be acidic pH 4. 7; chloroquine then cannot leave by diffusion. Antimalarial drugs are used for the treatment and prevention of malaria infection. Most antimalarial drugs target the erythrocytic stage of malaria infection, which is the phase of infection that causes symptomatic illness. The extent of preerythrocytic hepatic stage activity for most antimalarial drugs is not well characterized. Some of them, such as chloroquine, are more effective than quinine in suppressing the growth of the blood forms of the malarial parasite; others, such as primaquine, act upon both the blood and tissue stages of the parasite, thus producing complete cures and preventing relapses. All the newer antimalarials, unlike quinine, may be completely synthesized on a commercial scale.

Malaria is common in areas such as Africa, South America, and Southern Asia. Parasites that cause malaria typically enter the body through the bite of a mosquito.

Chloroquine has a high affinity for tissues of the parasite and is concentrated in its cytoplasm. As a weak base, it increases the pH of the intracellular lysosome and endosome. A more acidic medium in these organelles is needed for the parasite to affect mammalian cells. As a result, chloroquine inhibits growth and development of parasites.

  9. My doctor started me on plaquenil in combination with minocycline for my first round of treatment. I thought I had Levaquin, but I just had a look in my medicin box and it wasn't Levaquin, but Plaquenil (I think at some point my dr must have mentionned Levaquin, hence the confusion - and my bad memory played its role too).
